High-throughput automated protein expression analysis of human breast cancer HER-2 cell lines using 2D-DIGE MALDI-MS

Abstract

The use of 2D-DIGE MALDI-MS technology for high-throughput automated protein expression analysis of human breast cancer HER-2 lines was discussed. For the study two cell lines were obtained from ATCC and analyzed by two-dimensional differential in gel electrophoresis. The results from the global analysis of proteins in breast cell lines by 2D-DIGE indicate that up to 3.1% of proteins may be differentially expressed between HER-2 (+) and HER-2 (-) cells. It was suggested that out of the proteins differentially expressed, 13 appear to be co-expressed with HER-2, and 9 downregulated as a response to HER-2 overexpression.
